Turning on your PvP flag is instant, turning it off requires 5 minutes of non-PvP activity, or until you take a flight path or zone into an instance.

On PvP and RPPvP realms, you may only unflag in a same-faction zone.

On PvE and RP realms, you may unflag everywhere except in battlegrounds and opposite faction controlled areas.
